Job Description

  • Internship Experience (4 months)
  • Organization: National Center for Genetic Engineering and Biotechnology (BIOTEC, NSTDA)
  • Position: Research Intern
  • Responsibilities included Protein Expression and Purification.
  • Conducted recombinant protein expression in a bacterial system (E. coli) using IPTG induction.
  • Optimized protein expression conditions such as temperature (16–37°C), IPTG concentration, and induction duration to enhance yield and solubility of the protein.
  • Prepared and cultured strains using a shaking incubator and monitored growth by measuring OD600 (spectrophotometer).
  • Extracted proteins from cells using cell lysis methods such as sonication and centrifugation to separate soluble and insoluble fractions.
  • Purified proteins using affinity chromatography (His-tag) with Ni-NTA columns and controlled the process using gravity flow.
  • Prepared buffers and related solutions such as lysis buffer, wash buffer, and elution buffer while controlling pH levels.
  • Analyzed protein expression and purity using SDS-PAGE and gel staining (Coomassie Brilliant Blue).
  • Utilized basic laboratory equipment such as micropipettes, centrifuges, vortex mixers, autoclaves, and laminar flow hoods.
  • Performed work under aseptic technique and biosafety principles to minimize contamination risks.
  • Systematically recorded experimental results (lab notebook) and conducted preliminary data analysis.
